GeoMx Digital Spatial Profiler | Nanostring

CITOGEN offers those research centres and/or laboratories that want to use Nanostring technology, but do not have the equipment to analyze the samples, the possibility of doing it in our laboratory.

Once the project is known, our experts will help you choose the most appropriate panel for your study. Depending on the protocol to be applied, the samples will be processed in the suitable equipment and we will share the results for analysis.

The NanoString® GeoMx® platform makes Digital Spatial Profiling of gene expression possible.

What can it be used for?

NanoString’s GeoMx Digital Spatial Profiling (DSP) technology enables a highly multiplexed study of gene expression in selected regions of a slide, simultaneously.

Thus, the morphological context that can be obtained by immunohistochemical techniques is combined with the generation of complete transcriptomes and profiles of hundreds of protein analytes.

How does it work?

NanoString’s DSP technology combines standard immunofluorescence techniques with digital optical detection of molecular barcodes for digital spatial profiling of gene expression.

Oncology

The GeoMx® Cancer Transcriptome Atlas (CTA) is designed for comprehensive profiling of tumor biology, the tumor microenvironment, and the immune response. This panel allows you profiling the expression of over 1,800 genes covering around 100 pathways. It allows:

  • Profile the global immune response
  • Assess microenvironment immune activity
  • Quantify tumor reactivity to the immune response and therapeutic treatments
  • Profiling heterogeneous tumors, covering clinically derived gene sets (Tumor Inflammation Signature, PAM50…)

Furthermore, it can be customized with up to 60 additional targets of interest.

Immunology and Immuno-Oncology

The GeoMx Immune Pathways Panel is designed to profile key aspects of the tumor biology, tumor microenvironment and immune status. This panel allows you profiling 84 ARN targets covering several pathways. It allows:

  • Profile the global immune response
  • Assess microenvironment immune activity
  • Quantify tumor reactivity
  • Measure the 18-gene Tumor Inflammation Signature known to be associated with response to PD-1/PD-L1 inhibitor pathway blockade

Neuroscience

The GeoMx Whole Transcriptome Atlas (WTA) is designed for comprehensive profiling of celular biology, with full coverage of protein coding genes. It allows:

  • Spatial Profiling: Possibility of identifying the expression of genes in specific regions of the tissue, with mapping of specific pathways and functions, as well as the abundance of cell types.
  • Biomarker Discovery: Identification of biomarkers associated with a localization in the tissue, with the potential to identify processes or therapeutic targets not located with other types of assays on the complete tissue.
  • Pathway Analysis: The annotated content provides a spatial mapping of the activation of signaling pathways within discrete compartments of the tissue.

Protein Panels

Immunology e Immuno-Oncology

The Immune Cell Profiling Core allows 18 key immuno-oncology targets and immune cell markers (T cells, B cells, macrophages, NK cells, epithelium and stroma) to be analyzed with spatial resolution. Up to 7 additional modules (each with 10 targets) can be added to the “Core” module:

  • IO Drug Target Module: includes drug targets under development within the area of ​​immuno-oncology, including checkpoint molecules and metabolic mediators of immune function.
  • Immune Activation Status Module: includes additional checkpoint molecules that modulate T cell activation.
  • Immune Cell Typing Module: includes an expanded set of cell markers to further delineate the types of immune cells covered in the “Core” module, for example, T cell subtypes.
  • Pan-Tumor Module: includes an expanded set of markers to detect specific tumor types, including ER + / HER2 + breast tumors, hematopoietic neoplasms, and melanoma.
  • Cell Death Module: includes immunogenic and programmed cell death mediating proteins.
  • PI3K / AKT Signaling Module: includes key proteins involved in PI3K-AKT signal transduction and phosphorylated protein products that allow the activation of the pathway to be measured.
  • MAPK Signaling Module: it includes key proteins involved in MAPK signal transduction and phosphorylated protein products that allow measurement of pathway activation.

Neuroscience

The Neural Cell Profiling Core allows analyzing with spatial resolution 20 targets designed for the evaluation of the immune cell profile, to which can be added modules related to the profiling of neural cells, Alzheimer’s or Parkinson’s pathology, autophagy or subtyping of glia cells.
